5mC DNA methylation plays an important role in gene transcriptional regulation. Although BS-seq is a well-established method to detect DNA methylation, its harsh chemical treatment damages DNA and reduces the library complexity. TAPS (TET-assisted pyridine borane sequencing) enables us to detect 5mC DNA methylation accurately and sensitively, without affecting unmodified cytosine, which increases genome coverage and preserves library complexity. Currently, I am interested in applying this technique in single cell multi-omics to study cancer heterogeneity.
